Pathogenicity of three formulations of Beauveria bassiana and efficacy of autoinoculation devices and sterile fruit fly males for dissemination of conidia for the control of Ceratitis capitata

This study reports the pathogenicity of three formulations of Beauveria bassiana (Balsamo) Vuillemin (Hypocreales) – Bb-ET, GHA, and Bb-AES – and their application in panel-type and cylinder-type autoinoculation devices, and using sterile males as vectors for the control of Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) in coffee-producing areas of the Central Highlands of Guatemala. Using sterile adults of C. capitata under laboratory bioassays, mean lethal concentrations (LC50) of 1.4 9 107, 2.4 9 106, and 8.2 9 105 conidia ml-¹, and median survival times of 4.1 0.1, 4.2 0.1, and 3.8 0.1 days, were recorded for the strains GHA, Bb-AES, and Bb-ET, respectively. These values indicate that the three strains were sufficiently pathogenic and that their period of biological activity was considered adequate for using sterile flies as vectors of the inoculum. Observed percentages of sporulated wild C. capitata flies were 57.3, 44.7, and 44.3% for sterile fly vectors, panel devices, and cylinder devices, respectively. The total population reduction at the end of the study period was over 90% for the three treatments. Our results show that sterile males used as vectors or disseminator devices may represent a new control method for area-wide integrated management of Mediterranean fruit flies. The potential of this new approach and its integration with other control methods, including the sterile insect technique, is discussed.

The electrochemical generation of hydrogen peroxide with boron doped diamond electrodes was experimentally investigated to assess the influence of basic operating parameters on H2O2 production rate. Experiments were conducted in an undivided electrolytic cell, at room temperature, and different operating conditions of air bubbling, stirring and current density. Current density was the most influencing parameter affecting H2O2 generation. Therefore, an equation was proposed to describe hydrogen peroxide generation at BDD electrodes as a function of current density and electrolysis time. Experimental data showed that H2O2 concentration significantly increased during the first 45 min of electrolysis, and thereafter it remained constant until the end of the reaction. The equation was able to describe well this H2O2 production pattern. Additional experiments were conducted to validate the proposed equation. Good agreement between theoretical predictions and experimental data, as assessed by the Chi square goodness of fit test, was observed.

En este estudio se investigó la presencia de tres anti-inflamatorios no esteroideos (AINEs: naproxeno [NPX], ketoprofeno [KFN] y el ibupofreno [IBF], en cuerpos de agua superficial receptores de aguas residuales no tratadas de la ciudad de Tapachula (sur de México). Las muestras fueron tomadas durante tres campañas de muestreo en dos ríos (Coatán y Texcuyupan), una corriente urbana (Coatancito) y en una laguna costera (Barra San Simón). Las muestras se analizaron por cromatografía de gases-espectrometría de masas. Los resultados mostraron altas concentraciones de NPX (17.83 a 142.50 μg L-1), KFN (5.30 a 67.87 μg L-1) y IBF (3.97 a 31.30 μg L-1), presente en la mayoría de las muestras. El análisis de correlación entre los compuestos de interés y los parámetros fisicoquímicos muestran que las descargas de aguas residuales son las principales fuentes de estos contaminantes. Por otra parte, los índices de riesgo estimados sugieren una posible amenaza para los organismos acuáticos (algas, microcrustáceos y peces) debido a las altas concentraciones del NPX e IBF en aguas superficiales.

This study investigated the presence of three non-steroidal anti-inflammatory drugs (NSAIDs: naproxen [NPX], ketoprofen [KFN] and ibuprofen [IBF]), in surface water bodies receiving mostly untreated sewage from the city of Tapachula (South Mexico). Samples were taken during three sampling campaigns from two rivers (Coatán and Texcuyuapan), one urban stream (Coatancito), and one coastal lagoon (Barra San Simón), and the samples were analyzed by gas chromatography-mass spectrometry. Results showed high concentrations of NPX (17.83-142.50 μg L-1), KFN (5.30-67.87 μg L-1) and IBF (3.97-31.30 μg L-1), present in most samples. Correlation analysis between target compounds and physicochemical parameters supported the view that sewage discharge is the main source of these pollutants. Furthermore, estimated hazard indexes suggested a potential threat to aquatic organisms (algae, daphnids and fish) due to high concentrations of NPX and IBF in surface water.

Sterile Mediterranean fruit fly, Ceratitis capitata (Wied.), males were evaluated as vectors to spread Beauveria bassiana (Bals) conidia to wild C. capitata populations under field conditions. The inoculated sterile males were released by air, using the chilled adult technique over 7000 ha of coffee growing in Chimaltenango, Guatemala, Central America. The impact of releases was determined using dry traps baited with a food attractant. The effects of these releases on Apis mellifera, Linnaeus (honey bee), Hypothenemus hampei, Ferrari (coffee berry borer) and the parasitic mite Varroa destructor (Oudeman) were also evaluated. Inoculated sterile males were able to transmit fungal spores to 44% of the wild C. capitata flies captured in traps, which likely were infected through intra- and intersexual interactions during leks, mating or mating attempts. There was no transmission of the fungal spores to non-target insect species such as coffee berry borer, honey bees or varroa. We conclude that sterile males of Mediterranean fruit fly inoculated with B. bassiana can act as effective vectors of conidia to wild populations, constituting a safe, environmentally friendly and selective alternative for suppressing the medfly under a Sterile Insect Technique-based IPM approach.

In this study, we investigated the attraction of West Indian fruit fly, Anastrepha obliqua (Macquart) (Diptera: Tephritidae), to volatiles of three mango [Mangifera indica L. (Anacardiaceae)] cultivars in field cage tests. The number of flies captured with Multilure traps baited with Amate mature green mangoes was significantly higher than that captured in traps baited with Coche and Ataulfo fruits. There was no significant difference between the number of flies captured in traps baited with Coche or Ataulfo mangoes. Gas chromatography-mass spectrometry analysis of mango fruit volatiles identified 24, 22, and 19 compounds for Amate, Ataulfo, and Coche mango cultivars, respectively. A principal component analysis of the volatiles revealed that the Amate mango was more distant from the Ataulfo mango, and the latter cultivar was closer to the Coche mango. The compounds myrcene, α-pinene, β-selinene, and trans-β-ocimene were the most abundant in Amate mangoes, whereas 3-carene, β-selinene, terpinolene, and α-pinene were the predominant compounds of Ataulfo cultivars. In the Coche mango, the predominant compounds were 3-carene, β-selinene, terpinolene, and limonene. Traps baited with a blend of myrcene, α-pinene, and trans-β-ocimene captured more A. obliqua females and males than control traps. Flies were more attracted to the Super Q volatile extracts of Amate mango than to the three-component blend formulated in a ratio of 1:1:1. However, there was no significant difference between the number of flies caught by traps baited with Amate mango extracts and that caught by traps baited with the three-blend component when this was formulated according to the relative proportions in the mango extracts. Traps baited with myrcene, the major component, caught fewer flies than traps baited with Amate mango extracts.

En este trabajo presentamos resultados sobre el uso de trampas y feromonas comerciales para el monitoreo del gusano cogollero Spodoptera frugiperda, en cultivos de maíz y sorgo en la costa de Chiapas, México. El monitoreo del gusano cogollero con trampas con feromona es una herramienta poco explotada actualmente, que puede ser utilizada para determinar el momento preciso en que es necesario aplicar medidas de control en el cultivo. Después de varios años de ensayos, podemos decir que las trampas Scentry Heliothis y la Delta, cebadas con feromona Chemtica o Trece, proporcionaron las mejores capturas de machos de S. frugiperda, por lo que podrían ser utilizadas para el monitoreo de esta plaga en México.

In this work we present our results about the use of commercial traps and pheromones lures to monitoring the fall armyworm, Spodoptera frugiperda in maize and sorghum fields in the coastal of Chiapas, Mexico. The use of taps baited with pheromones is a tool that has scarcely explored to determine the optimal timing of control measures. After many years of trials, we have found that the Scentry Heliothis and delta traps baited with Chemtica or Trece pheromones gave higher caught of males of S. frugiperda, and it can be used to monitoring this pest in Mexico.
