Immunomodulatory effect of Lactobacillus casei in a murine model of colon carcinogenesis

We previously reported beneficial effects of the probiotic strain Lactobacillus casei 393 in hindering colon carcinogenesis in a 1,2-dimethylhydrazine (DMH)-induced BALB/c mouse model of colon cancer. In the present study, we investigated the effect of preventive administration of L. casei 393 on the levels of selected pro- and anti-inflammatory circulating cytokines, as well as subpopulations of splenic T cells. The resulting experimental data on IFNγ, TNFα, IL-10, and colon histological features demonstrated that administration of L. casei 2 weeks before DMH treatment impaired the pro-inflammatory effect of DMH, while maintaining the levels of the three cytokines as well as colon histology; it also modulated splenic CD4+, CD8+, and NK T cell subpopulations. The preventive administration of L. casei to DMH-treated mice increased IL-17A synthesis and Treg percentages, further indicating a tumor-protecting role. Together, the results suggest that the colon-cancer-protective properties of L. casei 393 involve the dampening of inflammation through cytokine homeostasis and the maintenance of a healthy T cell subpopulation dynamic. For these reasons, probiotics such as L. casei may contribute to the health of the host as they promote optimal control of the immune response. Further, they may be used as prophylactic agents in combination with standard therapies against colon cancer.

Evidence of the role that dietary carbohydrates (total carbohydrates, dietary fiber, total sugars, dietary glycemic index (GI) and glycemic load (GL)) exerts on insulin levels in adolescents is controversial. Thus, the aim of this study was to assess the association between dietary carbohydrates and insulin resistance in adolescents from Chiapas, México. A cross-sectional study was conducted in 217 adolescents. Sociodemographic, anthropometric, dietary and biochemical data were obtained. Total carbohydrates, dietary fiber, total sugars, dietary GI and GL were calculated from 24 h recalls. Two validated cut-off points for the homeostasis model assessment of insulin resistance (HOMA-IR) were used as surrogates of insulin resistance. Fasting insulin levels≥14.38 µU/mL were considered as abnormal. Multivariate logistic regression models were fitted to assess the association between tertiles of dietary carbohydrates and insulin resistance or hyperinsulinemia. In our study, adolescents with the highest dietary fiber intake had lower odds of HOMA-IR>2.97(OR=0.34; 95% CI:0.13–0.93) when adjusted for sex, age, body fat percentage and saturated fatty acids intake. No significant associations were found for the rest of the carbohydrate variables. In summary, high-fiber diets reduce the probability of insulin resistance in adolescents from marginalized areas of Chiapas, México.

Aedes aegypti y Aedes albopictus son especies de mosquitos transmisores del dengue, chikungunya y zika, graves enfermedades que entraron a México por zonas con condiciones desfavorables mezcladas con geografías y ambientes aptos para los zancudos. La participación comunitaria y la Técnica del Insecto Estéril pueden controlar estos mosquitos. Las experiencias al respecto en comunidades de Chiapas, Tabasco y Guatemala, se han destacado por una rica interacción entre pobladores y científicos.

The administration of probiotics is a promising approach to reduce the prevalence of colon cancer, a multifactorial disease, with hereditary factors, as well as environmental lifestyle-related risk factors. Biogenic polyamines, putrescine, spermidine, and spermine are small cationic molecules with great roles in cell proliferation and differentiation as well as regulation of gene expression. Ornithine decarboxylase is the first rate-limiting enzyme for polyamine synthesis, and upregulation of ornithine decarboxylase activity and polyamine metabolism has been associated with abnormal cell proliferation. This paper is focused on studying the protective role of Lactobacillus casei ATCC 393 in a chemically induced mouse model of colon carcinogenesis, directing our attention on aberrant crypt foci as preneoplastic markers, and on polyamine metabolism as a possible key player in carcinogenesis. BALB/c mice were administered 1,2-dimethylhydrazine dihydrochloride (DMH) to induce colon cancer (20 mg/kg body weight, subcutaneous, twice a week for 24 weeks). L. casei ATCC 393 was given orally (10 6 CFU, twice a week), 2 weeks before DMH administration. Hematoxylin and eosin staining, high-performance liquid chromatography, and Western blotting were used to evaluate aberrant crypt foci, urinary polyamines, and ornithine decarboxylase expression in the colon. The experimental data showed that the preventive administration of L. casei ATCC 393 may delay the onset of cancer as it significantly reduced the number of DMH-induced aberrant crypt foci, the levels of putrescine, and the expression of ornithine decarboxylase. Hence, this probiotic strain has a prospective role in protection against colon carcinogenesis, and its antimutagenic activity may be associated with the maintenance of polyamine metabolism.

La obesidad es un problema de salud pública a nivel mundial, se estima que el 52 % de los adultos y el 30 % de los niños sufren exceso de peso. Los mecanismos moleculares implicados en la obesidad permiten la comprensión de las enfermedades asociadas a ella como la hipertensión, enfermedades cardiometabólicas, eventos cerebrovasculares y resistencia a la insulina. Por una parte los eventos moleculares afectan las señales producidas por la insulina permitiendo su resistencia, por otra parte se induce un estado inflamatorio sostenido característico de las personas obesas, que están implicados en la formación de placas ateromatosas, estimulación de un estado protrombótico sostenido y aparición de eventos isquémicos. En estas enfermedades el estudio de las adipocinas como leptina adiponectina, factor de necrosis tumoral alfa, proteína c Reactiva, permite conocer el vínculo de la obesidad con las enfermedades crónicas además de ser un blanco de estudio activo para su utilidad diagnóstica.

Obesity is a public health problem worldwide. It has been estimated that 52 % adults and 30 % children are overweight. Knowledge about the molecular mechanisms involved in obesity leads to a better understanding of related conditions such as hypertension, cardiometabolic disease, cerebrovascular events and insulin resistance. Molecular events affect the signals issued by insulin, allowing resistance. A sustained inflammatory state is induced which is typical of obese people, characterized by the formation of atheromatous plaques, stimulation of a sustained prothrombotic state, and the occurrence of ischemic events. In the context of these conditions, the study of adipokines such as leptin, adiponectin, tumor necrosis factor alpha, C-reactive protein, serves to reveal the links between obesity and chronic disease, besides being a useful object of study to enhance diagnostic usefulness.
